If activated, the adhered monocytes will secrete inflammatory cytokines, such as monocyte chemoattractant protein-1 (MCP-1). After 48 hours of culture, the culture media was collected to measure the expression levels of various cytokines using ELISA. The following table shows the absorbance data of several MCP-1 standards of known concentrations, the media from monocytes cultured on the new polymeric coating, and the media from monocytes cultured on the stent without coating (control). Calculate the concentration of MCP-1 in the media from monocytes on the polymeric coating and control.

b. (12.5pts) You also completed ELISAs in the same manner to test the level of other inflammatory cytokines, interleukin-6 (IL-6), macrophage inflammatory protein-1α (MIP-1α) and macrophage inflammatory protein-1β (MIP-1β) as shown below. From the point of view of the inflammatory response, would you recommend continuing to develop this material for a next-generation DES? Justify your answer based on all of the ELISA data collected.

3. (10pts) A researcher would like to use a cage implant model system to look at the effect of the soluble fraction of an implant. He decides to use nickel to make the mesh. The holes in the mesh are approximately 1cm2. He then uses immunohistochemistry on the surrounding tissue implanted up to 4 weeks to investigate toxicity and the immune response. In a separate study, he determines that as the implant degrades, particles that are 100 microns2 are generated. Particles begin forming at 2 weeks. Do you trust the results from his cage implant model? List two reasons to support your answer.

(25pts) The pictures shown below are from an experiment in which osteoblasts (bone forming cells) were cultured on a poly (ethylene glycol)- based hydrogel material (designated as Acr-PEG-RGD) for 2 or 24 hours. This peptide sequence is known to bind to integrin receptors to promote cell attachment and spreading.
a. Describe two ways in which the RGD peptide could be linked to the surface of the material.
b. The above experiment was performed in the presence of a mixture of blood serum proteins in the cell culture medium. If twice as much of the same protein mixture were added to the culture medium, would cell attachment and spreading be affected for the no-RGD or the 5mM RGD samples? Explain your reasoning in both cases.
c. Explain why it appears that there is a larger number of cells in the 24-hour image than the 2-hour image for the 5-mM RGD sample.
